Scientists have demonstrated that lightly rubbing a frog's skin with a cotton swab can collect enough high-quality DNA to study endangered species without harming them. This breakthrough offers a new approach to conservation efforts in the Western Ghats and urban areas of India, where about half of all amphibian species are threatened with extinction.

Because this minimally invasive method proved effective for various species and sizes, the research group presented a foundation for conducting ethical wildlife studies. Previously, researchers often used invasive and painful methods, such as removing small pieces of amphibian fingers or tail tips to obtain tissue for DNA analysis.

Professor Gururaj KV, a faculty member at Srishti Manipal Institute of Arts, Design and Technology and author of the new study, noted that after long discussions, the team decided to try collecting samples from the skin instead of traditional methods like collecting tadpole tails, egg collection, finger clipping, or buccal swabs.

The new study was conducted by scientists from Srishti Manipal Institute of Arts, Design and Technology, Manipal Institute of Technology, and the Ashoka Foundation for Ecological Sciences and Environment. The work focused on 88 individuals from seven different groups of frogs and toads. The study showed that by applying 60 gentle touches with a cotton swab to the animal's back, abdomen, and limbs, scientists could collect enough detached skin cells to map the entire genetic code of the animal.

This new method is based on the fact that amphibians, like humans, constantly shed skin cells containing DNA. Scientists then use a process called Polymerase Chain Reaction (PCR) to repeatedly amplify or copy a specific gene, in this case the 16S rRNA gene, until there is enough material for sequencing. Sequencing allows reading the sequence of chemical letters A, T, C, G in the DNA, which helps identify new species, track diseases, and understand relationships between different populations.

The team found that over 86% of the swab samples successfully yielded DNA copies, and 67% were clean enough for sequencers to read. To ensure the practicality of the method for field researchers, the team tested various ways of storing and processing the swabs. They compared a professional laboratory kit with a cheaper, basic salt extraction method to see if they would get the same results.

It was also tested whether the swabs should be stored wet in alcohol or dry. Although both extraction methods yielded a similar amount of DNA, the professional kits provided cleaner samples. Interestingly, the researchers found that dry swabs actually provided higher purity than those stored in liquid, simplifying and reducing the cost of transporting and storing samples in remote tropical forests without the need for heavy bottles of chemicals.

This work is the first instance where the cotton swab sampling method has been thoroughly tested on multiple species of varying sizes in the complex, humid conditions of tropical India. While previous studies on very small frogs in other parts of the world sometimes faced difficulties obtaining sufficient DNA from swabs, this study succeeded even with tiny specimens, such as Microhyla genus frogs measuring 13 millimeters. Furthermore, researchers observed the animals after release and noticed that many frogs began croaking almost immediately, indicating that the process caused them significantly less stress compared to traditional methods.

Nevertheless, the researchers acknowledge that the effectiveness of the swab depends heavily on the texture of the frog's skin—whether it is smooth and slimy or dry and bumpy. For example, although the Malabar tree toad with bumpy skin yielded good results, the researchers noted that different skin types may require a different number of swab touches to achieve effectiveness. They also emphasized that while their method is excellent for species identification, more complex genetic studies may still require larger amounts of DNA obtainable from traditional tissue samples. Therefore, they recommend that future researchers conduct pilot studies on their target species to find the ideal balance.

Amphibians are key bioindicators because their condition reflects the health of the entire ecosystem. By providing an ethical, cost-effective, and reliable way to monitor these animals, this study allows scientists to gather vital data faster and more humanely. This aligns with the global 3R concept—Replacement, Reduction, and Refinement—which aims to make animal research more humane. Moreover, the new approach ensures that in the pursuit of saving endangered species, we no longer inflict unnecessary suffering on them, allowing us to protect nature while respecting the life within it.

A new study published in the journal Science Advances this Friday (31) suggests that seismic waves, similar to those generated by earthquakes, can be used to identify and map reserves of buried ice beneath the lunar surface.

The investigation was conducted by geologists affiliated with the University of Maryland (USA), the Lawrence Berkeley National Laboratory, and the University of Hawaii (USA). This work comes at a crucial time, as NASA's Artemis program plans crewed landings in the Moon's south polar region in 2028.

Water ice found in the deep, frozen shadows of the polar craters is considered a resource of great value for astronauts. When melted and purified, it can become potable water; when electrolyzed, it provides oxygen for breathing and hydrogen for rocket fuel. This implies that locating a constant supply of lunar ice could drastically reduce the amount of supplies future missions need to transport from Earth.

Currently, there is no precise knowledge about the quantity or exact location of ice on the Moon. Although satellites can scan the lunar surface from orbit, they can only visualize the superficial layer of the soil. Water ice deposits may be much deeper, and this is exactly the depth that the new seismic methodology aims to reach.

The theoretical basis of the research lies in a physical distinction: frozen soil and dry soil react differently when a seismic wave passes through them. Ice hardens the surrounding material, causing vibrations to propagate two to three times faster than in dry soil. Furthermore, ice-rich areas can cause the reflection of seismic energy instead of allowing its passage, a phenomenon comparable to the echo produced by sound hitting a wall.

Nicholas Schmerr, an associate professor in the Department of Geological, Environmental, and Planetary Sciences at the University of Maryland and a collaborator on the study, commented to Phys.org that a properly positioned seismometer on the Moon would have the capacity to detect such effects. He stated: 'We can use seismic waves not only to verify if ice is present but also to estimate approximately how much of it exists.'

Three methods to test the hypotheses

To confirm the proposed theories, the team implemented three distinct lines of work:

Harrison Lisabeth, a rock physicist at the Lawrence Berkeley National Laboratory, the study's lead author, and an alumnus of the University of Maryland, simulated the process by freezing an Arizona (USA) volcanic rock that, after being crushed, closely mimics lunar dust. He used X-rays to analyze how ice deposits in the small spaces between the soil grains.

Matthew Siegler, from the University of Hawaii and co-author, created detailed temperature maps of the Moon's south polar region, determining which craters maintained sufficiently low temperatures to preserve ice for billions of years.

Additionally, Schmerr, at the University of Maryland, conducted computational simulations of small lunar tremors, observing their propagation and interaction with subsurface ice. In all approaches, the ice left clear and measurable signs in the collected seismic data.

Scientific importance beyond practical use

Beyond the direct benefit to astronauts, lunar ice has significant scientific relevance. The shadowed craters on the Moon have the potential to freeze and trap volatile substances, such as water ice, preserving them without interference over vast periods. Since the lunar rocks themselves date back about four billion years, studying this ice could provide information on how water was distributed in the early solar system.

Schmerr emphasized: 'The Moon witnessed some of the most critical parts of the early solar system, including how water was delivered.' He added that 'Studying the ice deposited there could reveal how water spread and, ultimately, how Earth's oceans formed.'

The researchers' predictions will be tested in missions scheduled for 2026 and 2028. The Chinese Chang'e-7 mission, which will carry a seismometer, is expected to land near Shackleton Crater at the end of 2026, an area with several suspected ice deposits. In 2028, Artemis program astronauts may install the Lunar Environmental Monitoring Station, equipment that Schmerr helped develop for seismic exploration.

In conclusion, Schmerr stated: 'Our findings are laying the groundwork for observations we will make in the coming years.' He stressed: 'No one has physically measured the ice on the Moon yet, but now we have a prediction of what to observe. This is an important first step.'

OpenAI has identified new instances where autonomous artificial intelligence (AI) agents managed to escape containment environments during internal testing. This discovery expands an investigation that was already opened following the incident involving the Hugging Face platform, according to sources contacted by Reuters.

Two individuals familiar with the subject reported that these new events were detected during the company's public inquiry into how one of its agents managed to leave an environment that should have been isolated this month. According to one of the sources, the episodes are considered limited, and there is no evidence that the agents left OpenAI's internal network. The company is conducting an additional investigation into these new occurrences.

When reached, OpenAI directed attention to the note released on Tuesday (28), in which it mentioned analyzing 'broader activity of its models,' in addition to the Hugging Face intrusion.

Possible increase in pressure for regulation

Despite the new incidents being classified as restricted, this finding could intensify the demand for stricter standards for AI development. This occurs at a time when the topic has been receiving attention from both the White House and authorities in various countries.

Sources indicated that OpenAI expanded its investigation shortly before its main rival, Anthropic, admitted that the company's models were also responsible for a series of intrusions affecting three companies since April. Information about previous episodes involving OpenAI agents had not yet been made public.

Experts warn of risks

For AI safety experts, such cases reinforce the apprehension that large laboratories are developing autonomous agents with offensive capabilities at a pace faster than ensuring their control. Maurice Chiodo, a mathematician at the Centre for the Study of Existential Risk, University of Cambridge (England), told Reuters that there is an industry where professionals who create, develop, and release these tools are not keeping up with the responsibility of keeping them safe.

Reuters could not confirm the exact number of additional incidents or their dates. Three sources reported that OpenAI investigators and external experts are examining activity logs from past months to reconstruct the facts.

The Hugging Face case initiated the investigation

The investigation was started by OpenAI after the incident that occurred in early July, when a company agent infiltrated the Hugging Face infrastructure during an internal test designed to assess its behavior. According to OpenAI itself, the agent began acting unexpectedly for several days within the corporate network.

In this episode, four accounts belonging to four different companies were also breached. One of these companies was Modal, based in New York. Chiodo expressed great concern about the possibility that both OpenAI and Anthropic were not monitoring their agents in real-time while they were carrying out such actions.

Additionally, OpenAI only learned of the intrusion after Hugging Face managed to contain the event, alerted the FBI, and made the case public. OpenAI alleged that the report contained inaccuracies but did not specify which data was incorrect.

On Thursday (30), when releasing details about the incidents involving its own models, Anthropic acknowledged that real-time monitoring of evaluation logs could have facilitated earlier identification of the problem. For Chiodo, this demonstrates failures in supervising AI agents, as he commented: 'It seems like they weren't even looking.'
